AOD-9604 – the clean fat burner?
July 4, 2026
AOD-9604 is one of the rare scene peptides for which real human data exist: 6 clinical trials with almost 900 participants. The idea behind it is elegant, fat loss without a growth signal, but the large human trial missed its goal.
What it is about
At its core, AOD-9604 is the end piece of growth hormone, building blocks 176 to 191. It was developed in the 1990s at Monash University in Australia to use only the hormone’s fat-burning effect and leave out water retention, effects on blood sugar and the rise in IGF-1. In the lab and in overweight mice, this worked. The episode explains why this makes AOD almost the opposite of growth hormone boosters such as MK-677, and follows it up to the phase 2 trial in humans, in which weight loss was not significantly better than with placebo. What remains is a very good safety profile and an open question about the dosage form.
Key points
- A 2008 paper in the Journal of Endocrinology describes that the fragment increases fat burning without raising IGF-1 or activating the classic growth hormone signaling pathways.
- As the point of attack, the episode names the β3 receptor of fat cells, which starts fat breakdown and heat production; in overweight mice, weight and fat percentage fell.
- In the phase 2 trial in overweight people, AOD-9604 missed the primary endpoint, weight loss compared with placebo; in 2007 it was dropped as a drug candidate.
- The failed trial tested AOD as a tablet; because the digestive tract often breaks down peptides, too little active substance may have arrived, but a large trial with the injected form is lacking.
- In 6 clinical trials with almost 900 participants, AOD-9604 was practically as well tolerated as placebo, a rare data situation for a scene peptide.
- AOD-9604 was at the center of one of the biggest doping scandals in Australian sport, around the Essendon football club; its anti-doping status is considered disputed.
Where the data stop
Here data are not simply missing, here there is a result: the large human trial showed no significant difference from placebo in weight. The hope that the injected form performs better is plausible but unproven. The good tolerability says something about safety, not about efficacy; that AOD was classified as a dietary supplement ingredient in some countries mainly means that the safety framework fit.
In Germany, AOD-9604 is not an approved medicine, as of July 2026. What is sold loose in vials is not pharmaceutically tested; purity and actual quantity are not assured without analysis. For competitive athletes it is a delicate issue because of its disputed doping status.
