5-Amino-1MQ – fat-loss molecule fact-checked
July 4, 2026
5-Amino-1MQ has a reputation in the scene for burning fat, preserving muscle and providing energy, entirely without stimulants. Behind it is an elegant approach targeting the enzyme NNMT, which works impressively in mice and has never been tested in humans.
What it is about
5-Amino-1MQ is often sold as a peptide, but it is a small synthetic molecule that also works when swallowed. It slows the enzyme NNMT, nicotinamide N-methyltransferase. This enzyme consumes nicotinamide, a precursor of NAD+, and methyl groups; in obesity and in aged muscle it is overactive. The idea: if NNMT is inhibited, more NAD+ and more methyl reserve remain in fat and muscle cells, energy metabolism runs better, and less new fat is formed. The episode goes through the two central mouse studies, puts the enthusiastic user reports into context and asks what the permanent inhibition of an enzyme with normal tasks means. At the end stands a fascinating research approach against obesity and muscle wasting that still has the step to humans ahead of it.
Key points
- 5-Amino-1MQ is not a peptide but a small synthetic molecule from the group of NNMT inhibitors, and unlike many peptides it also works as a capsule.
- In cell experiments, inhibition of NNMT raised NAD+, new fat formation was curbed, and via the longevity switch SIRT1 energy metabolism ran better.
- In a 2018 mouse study, obese animals lost considerable fat and weight without eating less, solely through inhibition of the enzyme.
- In a second mouse study from 2019, the compound activated muscle stem cells in old animals after a muscle injury; the fibers became thicker, and strength was around 70 percent above the control.
- As a report: the community raves about fat loss, muscle gain and energy without a crash; these are user reports and protocols from the scene, not measurements.
- There are 0 studies in humans: no registered clinical trial and no approved product; all efficacy and safety data come from mice and cell culture.
Where the data stop
Everything known about efficacy and safety comes from mice and cell culture; every statement about humans is an extrapolation. The popular phrase “hardly any side effects” is also a claim, not a study finding. Also open is what broad, permanent inhibition does, because NNMT normally regulates the cell’s methyl group balance and redox equilibrium.
5-Amino-1MQ is not approved as a medicine in Germany, as of July 2026. It is sold on the gray market as a research chemical, without pharmaceutical quality control, with uncertain purity and quantity; some suppliers misleadingly declare it as a dietary supplement or peptide. It is not named on the World Anti-Doping Agency’s prohibited list, but as an unapproved substance with metabolic effects it very probably falls under the prohibited substances. Anyone who wants to support NAD+ and metabolism has proven levers in exercise, sleep and diet.
