Melanotan 2: the tanning peptide fact-checked

Melanotan 2 makes the skin turn brown without lying in the sun for hours. The peptide from the 1980s is the ancestor of no fewer than two approved drugs but never became one itself, and it carries a risk you need to know about.

What it is about

Melanotan 2 was developed at the University of Arizona, where researchers led by Victor Hruby and Mac Hadley wanted to prevent skin cancer by tanning the skin before sun exposure. It is a stable, ring-shaped copy of the hormone alpha-MSH and acts on several melanocortin receptors: in the skin on tanning, in the brain on appetite and desire. The episode tells how the sister peptide became a drug for a rare light-sensitivity disease and how a self-experiment led to a treatment for low sexual desire in women. It goes through the early human studies and dermatologists’ concerns about moles. What remains is probably the strongest known tanning signal, with a clear risk profile.

Where the data stop

The human data come from two small studies from the 1990s. The main problem is moles: Melanotan 2 can make existing ones darker or larger and cause new spots to appear; Langan and colleagues described this in case series in 2010. This makes early detection of melanoma more difficult. There are case reports of melanomas in users; a causal link has not been proven. Anyone with many or conspicuous moles or a family history of skin cancer should stay away from it; everyone else should have their skin examined beforehand and regularly.

Added to this are nausea, facial flushing and, rarely, a persistent erection, which is an emergency. A peptide tan does not replace sunscreen. Melanotan 2 is not approved as a medicine, and its sale as a tanning agent is prohibited in Germany and the EU, as of August 2026.
