Fasting & autophagy: the free upgrade fact-checked

The most powerful longevity tool of the series costs nothing: not eating. The episode takes apart the famous cellular mechanism behind it and checks what of it is really proven in humans — and what was extrapolated from mouse data.

What it is about

Autophagy literally means eating oneself and refers to the largest recycling program in biology: the cell packages damaged proteins and tired mitochondria, breaks them down and builds new things from the parts. The program is switched on by energy shortage, when the growth switch mTOR turns off and the energy sensor AMPK kicks in. The episode explains these mechanics, then critically examines the famous question of hours and sorts the fasting formats by what each of them delivers. After that come the athletic listeners’ worries about their muscles, the timing of the eating window, the question about women and the scene’s perennial question of what actually breaks a fast. At the end, fasting stands as a foundation, not as a substitute for everything else.

Addendum (as of October 6, 2026): A threshold in hours for autophagy has not been established in humans so far, not even beyond 24 hours. There is now a randomized study that measured autophagic flux under fasting, in blood cells: in 121 people with obesity, a difference from the control group appeared after 6 months of intermittent fasting, though in a post hoc analysis and without a comparison of different fasting durations (Bensalem 2025). Where the data stop

The most honest passage of the episode concerns measurability: in humans, autophagy can hardly be captured directly, because nobody looks continuously into the liver or brain, and blood markers are snapshots with pitfalls. The hour figures in circulation are therefore mostly invented or transferred. For the short formats, the cell-cleaning promise honestly remains hope plus mouse data; what is solidly supported there are the metabolic effects.

And fasting is not the right tool for everyone. Pregnant and breastfeeding women, children and adolescents, underweight people and people with a history of eating disorders are explicitly excluded. People with diabetes on medication need a medical adjustment because of the risk of hypoglycemia. Multi-day fasting belongs under medical supervision, and the refeeding afterwards is not a side issue but half the effect. Anyone who feels permanently weak, irritable or obsessive already has their body’s answer.
