Magnesium L-threonate: brain magnesium fact-checked

Everyone knows magnesium from the drugstore shelf, but ordinary supplements barely reach the brain. That is why a form intended to go exactly there was developed at MIT: magnesium L-threonate. The episode separates marketing from surprisingly solid science.

What it is about

Magnesium is involved in more than 600 enzyme reactions, and according to estimates about half the population does not reach the recommended intake. In the brain, it keeps watch at the NMDA receptor, the learning switch of the synapses, and ensures clean signals. However, the blood-brain barrier lets barely any additional magnesium from citrate, oxide or bisglycinate into the brain. The neuroscientist Guosong Liu and his team at MIT found a carrier in L-threonate, a derivative of vitamin C, that measurably raised magnesium levels in the brains of rats. The episode follows the path from rats to human studies on memory and sleep and classifies the common forms of magnesium. At the end stands a brain specialist that complements ordinary magnesium but does not replace it.

Key points

  • Slutsky and colleagues showed in Neuron in 2010 that rats given magnesium L-threonate learned faster and remembered better; old animals in some cases regained the learning performance of young ones.
  • In a randomized, double-blind study from 2016, cognitive performance improved after 12 weeks in adults around 60 with incipient memory problems, by a calculated equivalent of around 9 years of “cognitive age”.
  • A 2022 study with the further development Magtein PS found better memory and processing speed in healthy adults after just 30 days.
  • In a 2024 sleep study with 80 adults, 1 gram of threonate in the evening over 3 weeks brought more deep and REM sleep as well as more energy and better mood during the day.
  • 1 gram of threonate supplies only about 75 milligrams of elemental magnesium and does not cover daily requirements; basic supply works better via citrate or glycinate.
  • 1.5 to 2 grams of magnesium L-threonate daily are usual in the studies; less than 1.5 grams per daily dose is more of a fig-leaf addition.
  • As a report: users describe less rumination in the evening and a calmer mind; for the anti-stress effect in the body, cheaper magnesium also suffices.

Where the data stop

The human studies are still small, and the 2016 memory study was co-funded by the manufacturer, albeit properly randomized and double-blind. The effects are strongest in older people and those with poor magnesium status; young, well-supplied people who sleep well often notice little. Threonate is not an acute booster; the memory studies ran over weeks to months. For stress and mood, the evidence is more indirect.

Side effects are rare; at the start, some report mild head pressure or drowsiness. Anyone with kidney disease or taking medication should clarify magnesium with a doctor. Because the raw material is expensive, some products contain hardly any real L-threonate. And threonate can deepen 7 to 8 hours of sleep but cannot replace them.
