Klotho – the anti-aging protein from a mouse line gone wrong

A mouse line that was considered useless suddenly aged in fast motion and led to a gene that has since been named after the goddess of the thread of life. The famous 30 percent more lifespan is even true — it just belongs to a mouse.
What it is about
Klotho is produced in the kidney; a piece of it circulates in the blood and dampens insulin and growth factor signals. If the gene is missing, mice age prematurely; if it is turned up from conception onward, they live longer, and since 2025 there have for the first time been mice that were treated only as adults and lived longer. In humans there are gene variants, blood levels, a monkey experiment and reports from people who have a gene therapy administered in Honduras: less brain fog, waking up clearer. These are reports, collected by the provider and without a comparison group, not evidence of efficacy. At the end comes the most useful part of the episode: you can raise your own level without buying anything, with exercise, as measured in humans.
Key points
- In Barcelona in 2025, normally aging mice treated at 12 months of age lived 31.5 instead of 26.3 months; the authors themselves did not count the data from the females because of skin inflammation.
- Carriers of the KL-VS gene variant performed better in cognitive tests among 718 people in 2014, but slightly worse in a German study with 1,812 adults and among 1,480 very old people in Denmark.
- The blood level shows a U-shape: in Tuscany the lowest third had a roughly 78 percent higher risk of death, but in the US in 2025 the top fifth also had around 21 percent more than the middle.
- In 18 old rhesus monkeys in 2023, a single injection improved memory for 2 weeks; of 3 levels tested, only the lowest worked, and several authors hold shares in the company involved.
- From Honduras, treated people report less brain fog and faster thinking; the provider speaks of at least 60 hours of video testimonials, and the studies there had 14 people each and no placebo.
- A 2026 review of 30 studies with 2,765 people found a clear increase in the blood level through exercise, largest with strength training 3 times a week.
Where the data stop
To this day there is no completed, published placebo-controlled study in humans. The two studies in Honduras have 1 group, no placebo and no blinding; the one combined with follistatin has been completed since April 2026, and no results are in the registry. As of September 2026, a blinded study with 21 healthy people against placebo has been running on the same island since February 2026; results are expected by the end of the year. Nothing is approved, in any country in Europe. A gene therapy is built to stay; there is no off switch, and almost nothing has been collected on safety in humans.
A cream containing the supernatant of a cell culture in which Klotho is produced reduced sun damage by 48 percent in 16 adults over 16 weeks, without a control group and with the company’s CEO as a co-author; that is a finding about a skincare line, not about Klotho. And with exercise the marker does move, but many individual studies are weakly designed, and whether anyone lives longer because of it is open.
